After high school, Du Toit joined the Free State Cheetahs Academy and was the first-choice hooker for the Free State U19 side. Jacques was selected for the UFS Shimlas (2012) in the Varsity Cup competition. In 2013 he played for the Free State U21s and he was selected for the South African side which finished third in the 2013 IRB Junior World Championship in France.
He made his first-class debut for Free State XV playing off the bench in a Vodacom Cup victory over Kenyan invitational side Simba XV in 2013.
Du Toit played all 10 games in the 2016 Currie Cup, including the Semi Final against the Lions in Bloemfontein and in the Home Final against the Blue Bulls. (2016 Currie Cup Champions)
PRO14 debut – 1 September 2017 vs Ulster in Northern Ireland.
Du Toit, who was second in line after the Torsten van Jaarsveld in the past was set to be the starting hooker in 2018/19 however, a knee injury led to him missing the most of the season. He made his long-awaited return to the field during the tour of Ireland and Scarlets, which featured games against Connacht, Scarlets and Leinster.
